New Super-resolution microscope at the ALMF: Abbelight / Olympus Safe 360 with spectral demixing

The Abbelight / Olympus Safe 360 is our new super-resolution microscope for 2D- and 3D- multicolour SMLM (single molecule localization microscopy) at the ALMF. The Abbelight Safe 360 module is connected to an Olympus IX83 microscope equipped with a 100x / NA 1.5 oil immersion objective. The Abbelight Safe360 module is coupled to four laser lines (405, 488, 532 and 640nm) and two cameras (Hamamatsu Fusion) for fast multicolour super-resolution imaging (e.g. STORM and PALM). In addition to all conventional SMLM modes, it also offers spectral demixing, which allows detecting up to three colours with inherent chromatic alignment of the channels and therefore offering simpler and more precise super-resolution imaging of multiple targets. Imaging is possible with Epi-, HiLo- and TIRF-illumination.

Temperature control of the sample is possible between room temperature and 42°C by directly heating the coverslip using VAHEAT from Interherence.
