The development of transcriptome-wide RNA localization screens, together with the more recent advent of scRNA and spatial sequencing approaches, have revealed that RNA expression, processing and distribution are tightly regulated in space and time across species and cell types. These discoveries, performed in a range of biological systems (from cultured cells to organoids and model organisms), underscore the need to detect transcripts with both cellular and subcellular resolution and to quantitatively analyze their abundance and localization.
This course will provide comprehensive theoretical and practical training covering all key steps and the latest developments in quantitative FISH methods, while also offering a broad perspective on their applications to a wide range of biological questions.
Scientific talks by leaders in the field will additionally highlight application of ISH techniques in diverse biological models and introduce complementary methods to study RNA dynamics (e.g. localization, localized translation), along with transcriptome-wide and super-resolution approaches.
Through dedicated networking activities, project presentations and interactive discussion sessions, participants will engage with a vibrant community of biologists working on spatially resolved RNA imaging.
In this course, participants will learn how to design, perform and quantitatively analyze in situ hybridization (ISH) experiments to detect specific RNA molecules and study their processing in a variety of biological samples. They will receive hands-on training in complementary methods, including single-molecule FISH and hybridization chain reaction.
Practical modules will teach participants how to design robust, specific probes for diverse applications, how to fluorescently label them and how to use them for reliable and reproducible detection. Dedicated imaging and image analysis sessions will provide a solid foundation in best practices as well as knowledge on community-standard softwares.
