Capturing mRNA translation dynamics is critical for understanding gene expression across diverse biological contexts. Translation changes are often the first response when cells encounter environmental or physiological stress, and shifts in ribosome occupancy across transcripts, elongation speed, or initiation site can rapidly remodel the proteome before transcriptional reprogramming.
Capturing these rapid, codon‑level changes is essential to define how cells sense
damage, activate the integrated and ribotoxic stress responses, and prioritize synthesis of protective factors. Ribosome profiling approaches are currently the only methodologies that can measure transcriptome‑wide translation dynamics in vivo, at codon resolution.
The course is aimed at PhD students and early post-doctoral researchers who are entering the field of posttranscriptional regulation of gene expression.
Participants will learn a robust ribosome profiling protocol and how to adapt it to
diverse experimental systems, including applications that detect ribosome collisions as reporters of a ribotoxic stress response. They will also be trained in core computational analysis and quality control, using new resources including RiboSeq Data Portal and RiboCrypt.
By engaging leading developers of ribosome profiling strategies, the course will provide an experimental entry point to this demanding field, with practices, and reporting standards, and will help grow a global community of advanced users.
