They provide a transformative shift in how we understand cellular functions and the behaviour of proteins in their natural environments,\u201d said <a href=\"https:\/\/www.ebi.ac.uk\/people\/person\/sameer-velankar\/\">Sameer Velankar, Team Leader and Senior Scientist at EMBL-EBI<\/a>. \u201cThis collection is an invaluable resource for the scientific community that will help enrich our understanding of cellular mechanisms.&#8221;<\/p>\n\n\n\n<article class=\"vf-card vf-card--brand vf-card--striped vf-u-margin__bottom--800\" default>\n  <div class=\"vf-card__content | vf-stack vf-stack--400\">\n      <h3 class=\"vf-card__heading\">\n      What is cryo-ET?    <\/h3>\n                <p class=\"vf-card__text\"><span style=\"font-weight: 400;\">Cryo-ET involves flash-freezing biological specimens to capture them in their natural state, avoiding the distortion or damage that traditional preparation methods might cause. This technique then uses a beam of electrons to image the sample, creating a series of two-dimensional images from different angles. These images are reconstructed into detailed three-dimensional models, offering insights into the architecture of cells at the level of individual proteins without the need for crystallisation, allowing scientists to visualise proteins in their natural state and cellular context.<\/span><\/p>\n      <\/div>\n<\/article>\n\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Exploring the microscopic universe<\/strong><\/h2>\n\n\n\n<p>Cryo-ET allows scientists to visualise proteins in their cellular environment in 3D, bridging the gap between structural and cellular biology. Today, over 1,800 reconstructed tomograms and associated raw data of <em>Chlamydomonas reinhardtii<\/em> have been made freely available in the EMPIAR database helping researchers to investigate the mechanisms behind cellular functions and dysfunctions. Access to these data lays the groundwork for advancements and new tool development in disease treatment, drug development, and more.<\/p>\n\n\n\n<p>\u201cBy making such a large cryo-ET dataset freely accessible via EMPIAR, we don&#8217;t just aim to offer a glimpse into cellular complexity; but rather to help unveil a new frontier in structural biology. This dataset, a collaborative effort led by Thermo Fisher Scientific and academic research groups, signifies a paradigm shift in how we visualise proteins in their natural environments. The open accessibility of this dataset via EMPIAR database is designed to significantly benefit cryo-EM, image processing, and machine learning communities, helping drive advancements in algorithm development and data analysis,\u201d said Abhay Kotecha, Group Leader, Applications Scientists at Thermo Fisher Scientific. \u201cAs cryo-ET continues to evolve, so too does our ability to explore the complexities of life, driven by a commitment to open access and collaborative research.\u201d<\/p>\n\n\n\n<p><em>Chlamydomonas reinhardtii<\/em>, the organism used to create these image data, is a unicellular green alga used as an exemplary model for cryo-ET. Its unique cellular architecture, featuring both photosynthetic subcellular structures and standard eukaryotic organelles, makes it a versatile subject for exploring the complexities of cellular processes at an ultrastructural level.&nbsp;<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Navigating the data&nbsp;&nbsp;<\/strong><\/h2>\n\n\n\n<p>Alongside these new data, EMPIAR has introduced a new prototype tomogram browser for the visualisation of cryo-ET data. Designed with a user-friendly interface, this tool allows scientists to navigate and visualise high-resolution images of cellular structures with ease.
