{"id":6185,"date":"2015-11-30T15:15:29","date_gmt":"2015-11-30T14:15:29","guid":{"rendered":"http:\/\/news.embl.de\/?p=6185"},"modified":"2024-04-19T15:49:11","modified_gmt":"2024-04-19T13:49:11","slug":"1511-paulsen","status":"publish","type":"post","link":"https:\/\/www.embl.org\/news\/lab-matters\/1511-paulsen\/","title":{"rendered":"Welcome: Malte Paulsen"},"content":{"rendered":"\n<p>The <a href=\"http:\/\/www.embl.de\/services\/core_facilities\/flow_cytometry\" target=\"_blank\" rel=\"noopener noreferrer\">Flow Cytometry Core Facility<\/a> uses powerful lasers to analyse and sort cells as they stream very rapidly past sensitive detectors. The technique\u2019s ability to process <a href=\"http:\/\/news.embl.de\/science\/1410_flow\/\" target=\"_blank\" rel=\"noopener noreferrer\">thousands of cells in seconds<\/a> enables scientists to \u2018fish\u2019 rare cells out of a sample so they can then grow colonies of those cells in a lab dish, sequence their genomes or analyse other features of interest.<\/p>\n\n\n\n<blockquote class=\"vf-blockquote\"><p><strong>&nbsp;<\/strong>There\u2019s nothing nicer than seeing a user leave the facility happy \u2013 that\u2019s the pay<\/p><\/blockquote>\n\n\n\n<h3 class=\"wp-block-heading\">What got you interested in science in the first place?<\/h3>\n\n\n\n<p>When I was still little, my father, a chemist, used to show me and my brother small things with our children\u2019s chemistry set. It was especially cool around New Year\u2019s Eve when we tried to build some firecrackers on our own \u2013 that got me interested in science! Later, I admit I found pure chemistry slightly boring, so I took up biology and then biochemistry instead.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Name one tool you can\u2019t do without<\/h3>\n\n\n\n<p>I can\u2019t do without coffee\u2026 but as a tool: lasers and good optics. It is the mix of different lasers and very precisely matched detection optics that allow us to catch even the dimmest signal to sort samples. That ability is sometimes very important for research here at EMBL and we aim to customise our approach as much as possible to prepare the instrument optimally for each user\u2019s assay.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Who are your main users? What types of projects do you mostly support?<\/h3>\n\n\n\n<p>Our user base is very broad. This is quite interesting here at EMBL, actually: we have users from every Unit. We do a lot of cell sorting from cell cultures of course, but we\u2019re also seeing more and more samples coming from fish, a lot of fruit fly cells, and \u2018fancier\u2019 organisms. Flow cytometry at EMBL is not the typical flow cytometry set-up like in clinics, where you basically have your T-cells, your B-cells, your blood and maybe some lavages; here, it\u2019s more biochemistry and cell biology. You get a lot of clone lines from genome editing \u2013 we select those out based on fluorescent markers and purify them by single-cell sorting so users can make colonies from those cells. We also have a lot of preparatory sorts, where people say \u2018we need a couple of specific cells to do downstream analysis\u2019, like sequencing. We\u2019re usually at the start or an intermediate step for a project. Very often we\u2019re not the last step \u2013 we have to be extremely good at our job, because there\u2019s a lot of work that comes afterwards and a lot of work before, so we really have to be up to the task on the day.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">What is your philosophy for running your facility?<\/h3>\n\n\n\n<p>The user has to be happy, he or she is the top priority: their project, their sample \u2013 we\u2019ll do everything to make it possible that it runs. What really drives me is that there\u2019s nothing nicer than seeing a user leave the facility happy \u2013 that\u2019s actually the pay. If you know, \u2018I\u2019ve done it, they\u2019ve got what they came for, they didn\u2019t leave empty-handed\u2019 \u2013 this is the best. It\u2019s what I hope for every morning when I come to work. The same way, if something doesn\u2019t work, I take that on board, too. It\u2019s not like we\u2019re not in the lab anymore: in core facilities it is quite common that we experience every user\u2019s day.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">If there was a motto hanging at your facility\u2019s door, what would it be?<\/h3>\n\n\n\n<p>\u2018Got questions? Got interesting ideas? Want to do something? Just knock and come in.\u2019 This is what core facilities are about: we need to be approachable. If the user has an idea and doesn\u2019t mind walking over and just knocking on the door, that\u2019s perfect. The more they come with ideas, questions and discussions, the better. Once we know what to read up on, we can start understanding if we, with our tools, can do a task, or if we need something else or adapt our current setup to get the job done.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Are there new avenues you\u2019d like to explore in your facility?<\/h3>\n\n\n\n<p>We have to look closely at what is hot in technology \u2013 at the moment there\u2019s a couple of things happening, but we have to see what is really useful. And we have to see what our users need and wish for in addition to what we offer and wish for. I\u2019ve been writing emails and meeting with users, and I\u2019ve had nice feedback \u2013 responsiveness is really good here. EMBL is a great place to be if you have ideas that will benefit the science.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">What\u2019s the best advice you\u2019ve received (or wished you had received) in your career?<\/h3>\n\n\n\n<p>I\u2019m now doing what I like partly because my PhD advisor <a href=\"http:\/\/www.dkfz.de\/en\/mol_embryology\/\" target=\"_blank\" rel=\"noopener noreferrer\">Christof Niehrs<\/a> \u2013 who actually did his PhD at EMBL&nbsp;\u2013 said \u2018you know what, you\u2019re a helpful character, why not take your skills into core facilities?\u2019 He supported me with this and introduced me to a really good mentor, Bernhard Korn, who then showed me a lot about how core facilities work and how you should organise your core facility \u2013 I took this to heart. When I got the chance to go for a core facility, I said \u2018OK, let\u2019s give it a try,\u2019 and from day one I was happy with that decision. It\u2019s still my thing.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Why do you do what you do?<\/h3>\n\n\n\n<p>Because I love it! I really like my job, and I really love the technology \u2013 I think the mix of science and technology in flow cytometry is really cool. It\u2019s still a very evolving field \u2013 it\u2019s not stale.
