Determining how often, fast, and strongly these interactions happen is key to understanding how cells work on a molecular level. This is where biophysics comes into play.<\/p>\n\n\n\n<p>EMBL Hamburg\u2019s <a href=\"https:\/\/www.embl.org\/groups\/sample-preparation-characterisation\/\">Sample Preparation and Characterisation (SPC) Facility<\/a> offers scientists access to almost all available biophysics technologies. With the help and advice of the expert staff, users can perform precise measurements and data analysis.<\/p>\n\n\n\n<p>Located at the <a href=\"https:\/\/www.cssb-hamburg.de\/\">Centre for Structural Systems Biology (CSSB)<\/a>, the facility is just a few meters away from the EMBL beamlines for <a href=\"https:\/\/www.embl.org\/groups\/macromolecular-crystallography\/\">crystallography<\/a> and <a href=\"https:\/\/www.embl.org\/groups\/small-angle-x-ray-scattering\/\">SAXS<\/a>, and the CSSB\u2019s cryo-EM facility. Being able to prepare samples and conduct X-ray or cryo-EM experiments during a single visit gives the users the flexibility to quickly adjust their sample preparation protocol for their measurements.<\/p>\n\n\n\n<p>Users can also send their samples to EMBL Hamburg to use the SPC Facility and the EMBL beamlines remotely.<\/p>\n\n\n\n<p>Mar\u00eda Garc\u00eda Alai, the Head of EMBL\u2019s SPC Facility, emphasises providing reliable and excellent quality data. The facility staff ensure that all instruments are perfectly calibrated, and the sample preparation process is tailored to the needs of each user.<\/p>\n\n\n\n<p>Besides helping users conduct measurements, the SPC Facility provides support in analysing data. In 2021, <a href=\"https:\/\/www.embl.org\/news\/science\/biophysics-analysis-made-easy-with-an-online-tool\/\">they released the eSPC platform<\/a>, where users can analyse data from biophysics experiments, for free and from home.<\/p>\n\n\n\n<p>The SPC Facility is part of several European transnational access programs, such as iNEXT-Discovery, <a href=\"https:\/\/www.embl.org\/news\/lab-matters\/instruct-centre\/\">Instruct-ERIC<\/a>, and <a href=\"https:\/\/www.embl.org\/news\/lab-matters\/spc-mosbri\/\">MOSBRI<\/a>, which facilitate scientists\u2019 access to research infrastructures. The facility welcomes users from different backgrounds and disciplines, and also offers training in cutting-edge biophysics techniques.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\" id=\"A1\"><strong>From crystallisation screens to mass photometry<\/strong><\/h2>\n\n\n\n<p>Mary Spiliopoulou from the University of Patras in Greece sent her samples to the SPC Facility for a crystallisation screen. The facility\u2019s automated platform for high-throughput crystallisation allowed her to obtain crystals for an X-ray experiment at the EMBL beamline P13.<\/p>\n\n\n\n<blockquote class=\"vf-blockquote | vf-stack vf-stack--400\">\n    <p class=\"vf-blockquote__text\">\u201cI used the knowledge of the SPC Facility staff and then the diffraction experiments were very easy,\u201d said Spiliopoulou.<\/p>\n\n\n<\/blockquote>\n\n\n\n<div style=\"height:30px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<p>Katharina Weinh\u00e4upl from the University of Porto in Portugal used the SPC Facility several times, remotely and in person, to prepare her samples for a cryo-EM experiment. Among other techniques, she found the detergent screen particularly helpful.<\/p>\n\n\n\n<blockquote class=\"vf-blockquote | vf-stack vf-stack--400\">\n    <p class=\"vf-blockquote__text\">\u201cWhat made a difference for me is that you can use the SPC Facility remotely,\u201d said Weinh\u00e4upl. \u201cYou just send your samples, and you will get your results very fast.\u201d<\/p>\n\n\n<\/blockquote>\n\n\n\n<div style=\"height:24px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<p>Jacek Plewka from the Jagiellonian University in Poland came to the SPC Facility to conduct mass photometry experiments for his work on SARS-CoV-2. This new technique enabled him to directly detect the interaction between two viral proteins at very low concentrations, in near-physiological conditions. Later, he analysed the data using the eSPC platform.<\/p>\n\n\n\n<blockquote class=\"vf-blockquote | vf-stack vf-stack--400\">\n    <p class=\"vf-blockquote__text\">\u201cIf it weren\u2019t for the SPC, we would not be able to do some of the measurements because the equipment would be extremely expensive for us,\u201d said Plewka.<\/p>\n\n\n<\/blockquote>\n\n\n\n<div style=\"height:23px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n","protected":false},"excerpt":{"rendered":"<p>EMBL Hamburg\u2019s Sample Preparation and Characterisation (SPC) Facility offers scientists access to almost all available biophysics technologies.
