{"id":5066,"date":"2015-08-26T11:21:12","date_gmt":"2015-08-26T09:21:12","guid":{"rendered":"http:\/\/news.embl.de\/?p=5066"},"modified":"2024-04-19T15:15:08","modified_gmt":"2024-04-19T13:15:08","slug":"1508_super-impressions","status":"publish","type":"post","link":"https:\/\/www.embl.org\/news\/events\/1508_super-impressions\/","title":{"rendered":"Super impressions"},"content":{"rendered":"\n<p>Super-resolution microscopy pushes the boundaries of conventional microscopes, enabling scientists to visualise&nbsp;structures with higher resolution than ever before, increasing&nbsp;our understanding of important molecular pathways inside living cells \u2013 a feat that earned its developers the <a href=\"http:\/\/news.embl.de\/science\/1410_hell\/\" target=\"_blank\" rel=\"noopener noreferrer\">2014 Nobel Prize in Chemistry<\/a>.<\/p>\n\n\n\n<p>Over six days, course participants covered theory and&nbsp;practical applications, gaining hands-on experience with the latest&nbsp;commercially available <a href=\"http:\/\/www.leica-microsystems.com\/products\/super-resolution-microscopes\" target=\"_blank\" rel=\"noopener noreferrer\">super-resolution microscopes<\/a> for STED and GSDIM localisation, alongside in-depth tuition and talks from&nbsp;more than a dozen renowned guest researchers and&nbsp;experts from EMBL and Leica Microsystems. Towards the end of their busy week, we asked a few of the participants for their impressions.<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">What\u2019s one thing you\u2019ll definitely be taking back with you to your lab?<\/h3>\n\n\n\n<p>\u201cThere are certainly some techniques that I really wanted to try, and here I got to ask about technical details and see how things work in practice. I came in with my particular set of preferences, what I liked and didn\u2019t like, but now this course is changing my mind, or widening my perspectives.\u201d <strong>Vaclav Beranek, MRC Laboratory of Molecular Biology, UK<\/strong><\/p>\n\n\n\n<blockquote class=\"vf-blockquote\"><p>This course is changing my mind, or widening my perspectives.<\/p><\/blockquote>\n\n\n\n<p>\u201cThere\u2019s one fluorophore that I could never get to work in my lab, labelling kidney tissues. I got a great tip that you should use really, really low depletion power for that fluorophore \u2013 now I know it wasn\u2019t the fluorophore itself, it was us, and I can go back and try it again.\u201d <strong>David Unnersjoe-Jess, Royal Institute of Technology, Sweden<\/strong><\/p>\n\n\n\n<p>\u201cThe teaching, particularly the microscope sessions. Normally someone shows you once during training, and then you\u2019re on your own \u2013 the first time you use the microscope, you\u2019re looking around, trying to remember \u2018where did they click?\u2019. Here, with the trainer beside you, you work in small groups and gain a lot more confidence on the system. Then we could use all of these tools they have developed.\u201d <strong>Vaclav Beranek<\/strong><\/p>\n\n\n\n<p>\u201cI wish that our institute had something like the Advanced Light Microscopy Core Facility. The set up here is amazing \u2013 to have it in house, buzzing with people who can help and give you cool ideas. My wish list: Marko Lampe and his microscopes!\u201d <strong>Ella Fung<\/strong><\/p>\n\n\n\n<h3 class=\"wp-block-heading\">What was the coolest application you learned of?<\/h3>\n\n\n\n<p>\u201cThere was a talk by Katrin Willig about a really cool application of STED microscopy, where they actually <a href=\"http:\/\/www.leica-microsystems.com\/science-lab\/sharp-live-images-from-the-mouse-brain\/\">looked at the brain of a living mouse<\/a>. I was really impressed by how deep they could go and still keep the resolution.\u201d <strong>David Unnersjoe-Jess<\/strong><\/p>\n\n\n\n<blockquote class=\"vf-blockquote\"><p>As a PhD student, I could see what was achievable.<\/p><\/blockquote>\n\n\n\n<p>\u201cI really liked what Christian Eggeling has done, because I think he uses the technique where it provides really interesting new biological knowledge and not just beautiful images. <a href=\"http:\/\/www.leica-microsystems.com\/science-lab\/a-straightforward-approach-for-gated-sted-fcs-to-investigate-lipid-membrane-dynamics\/\" target=\"_blank\" rel=\"noopener noreferrer\">Eggeling studies lipids with these super-resolution techniques<\/a>,&nbsp;and it seems like a really powerful tool for that.\u201d <strong>Vaclav Beranek<\/strong><\/p>\n\n\n\n<p><span style=\"line-height: 1.428571429;\">\u201cGroup leader Jan Ellenberg presented <\/span><a style=\"line-height: 1.428571429;\" href=\"http:\/\/www.embl.de\/aboutus\/communication_outreach\/media_relations\/2013\/130711_Heidelberg\/index.html\" target=\"_blank\" rel=\"noopener noreferrer\">a PhD student\u2019s project<\/a>&nbsp;using these techniques to visualize the structure of a molecular complex.
