{"id":47520,"date":"2022-03-16T09:43:32","date_gmt":"2022-03-16T08:43:32","guid":{"rendered":"https:\/\/www.embl.org\/news\/?p=47520"},"modified":"2024-03-22T15:27:28","modified_gmt":"2024-03-22T14:27:28","slug":"putting-cryptococcus-in-context","status":"publish","type":"post","link":"https:\/\/www.embl.org\/news\/science\/putting-cryptococcus-in-context\/","title":{"rendered":"Putting Cryptococcus in context"},"content":{"rendered":"\n<p>Found all over the world, the fungi <em>cryptococcus<\/em> usually doesn\u2019t affect healthy individuals.&nbsp; However, it sometimes causes severe meningitis in immunocompromised individuals, like people with HIV. Infection usually occurs when people inhale the fungi, spreading it to the lungs and brain, possibly with fatal consequences.<\/p>\n\n\n\n<p>Greetje Vande Velde, Research Professor at KU Leuven, has studied <em>cryptococcus<\/em> for almost a decade. How the fungi are able to cross the blood brain barrier is still unknown, and that\u2019s why Vande Velde and Jim Swoger, Head of the <a href=\"https:\/\/www.embl.org\/groups\/mesoscopic-imaging-facility\/\">Mesoscopic Imaging Facility<\/a> at EMBL Barcelona, started collaborating several years ago.<\/p>\n\n\n\n<p>In 2015, Vande Velde was a postdoc at KU Leuven and Swoger still worked in the Sharpe Lab, at the <a href=\"https:\/\/www.crg.eu\/\">Centre for Genomic Regulation<\/a> (CRG) in Barcelona. She spent a research fellowship in the Sharpe lab to visualise mouse organs in 3D with Swoger, using the recently invented techniques Optical Projection Tomography (OPT) and Selective Plane Illumination Microscopy (SPIM).<\/p>\n\n\n\n<p>\u201cGreetje\u2019s expertise is in 3D <em>in vivo<\/em> imaging, which allows for live observation of disease spread, but it doesn\u2019t allow us to look at cells themselves within the context of organs and tissues,\u201d Swoger said. \u201cOPT and SPIM allow us to image complete mouse organs <em>ex vivo<\/em> at high resolution in 3D and help us see the spatial organisation of the cryptococci.\u201d<\/p>\n\n\n\n<div class=\"vf-embed vf-embed--16x9 | vf-u-margin__bottom--400\">\n<iframe src=\"https:\/\/www.youtube.com\/embed\/U1gfpJ-qesc\" frameborder=\"0\" controls allowfullscreen><\/iframe><\/div>\n\n  \n<figcaption class=\"vf-figure__caption vf-u-margin__top--200\">Greetje Vande Velde from the KU Leuven in Belgium and Jim Swoger, Head of the Mesoscopic Imaging Facility at EMBL Barcelona, explain how their collaboration enabled them to observe the 3D organisation of new <em>cryptococcus<\/em> lesions in infected brains.<\/figcaption>\n\n\n\n\n\n<p>In 2017, <a href=\"https:\/\/www.embl.org\/sites\/barcelona\/\">EMBL Barcelona<\/a> and the Mesoscopic Imaging Facility (MIF) were established, with Jim Swoger as Head of MIF. The project then went a step further by using a combination of the two imaging technologies used at the CRG: OPT plus SPIM. This became the hybrid OPTiSPIM.&nbsp; SPIM provided high resolution, which complemented the imaging speed and multi-modality (fluorescent and absorbing contrasts) of OPT.<\/p>\n\n\n\n<p>Vande Velde and Swoger imaged entire mouse lungs and brains infected with <em>cryptococci<\/em> to get 3D images at cellular resolution. They could then observe the fungi in the context of entire organs and see how fungal populations were organised in the folds of the brain.<\/p>\n\n\n\n<p>These imaging technologies and the sample preparation protocols allowed the researchers to make novel observations on where and how the fungi assembled \u2013 something that hadn\u2019t been captured previously. <em>Cryptococcus<\/em> populations were generally organised around the microvasculature of the brain, which is the system of tiny blood vessels like capillaries and venules. Such observations are pivotal to further probing how the fungi enter and infect the brain.<\/p>\n\n\n\n<p>\u201cThe collaboration with the MIF helped us achieve important insights towards understanding the disease pathogenesis.\u201d Vande Velde said. \u201cThere have been cases of apparently healthy individuals with severe cryptococcal infections that challenge our understanding of this disease to this day.
