{"id":2791,"date":"2014-11-28T13:26:36","date_gmt":"2014-11-28T12:26:36","guid":{"rendered":"http:\/\/news.embl.de\/?p=2791"},"modified":"2025-07-14T12:45:36","modified_gmt":"2025-07-14T10:45:36","slug":"1411_massif","status":"publish","type":"post","link":"https:\/\/www.embl.org\/news\/science\/1411_massif\/","title":{"rendered":"MASSIF step forward"},"content":{"rendered":"\n<h3 class=\"wp-block-heading\">By Matthew Bowler<\/h3>\n\n\n\n<p>MASSIF1, operated by the <a href=\"https:\/\/www.embl.org\/groups\/mccarthy\/\">EMBL\/ESRF Joint Structural Biology Group<\/a>, offers a unique, fully automated service for sample evaluation and data collection from crystals of macromolecules. The new service is not designed to replace all user visits to the synchrotron but rather to do the hard work of screening crystals through the night, freeing researchers to spend time on more challenging data collection problems and study the underlying biology.<\/p>\n\n\n\n<p>Users interact with the beamline by describing experimental requirements that are used by the beamline software to set data collection parameters and results are viewed via a database. Beam time is booked flexibly and samples then enter a queuing system. The service is made possible by RoboDiff \u2013 a new ESRF-developed sample changer that also acts as a goniometer \u2013, a highly intense X-ray beam and complex workflows that fully evaluate samples, identify and centre on the best parts of the crystal, and collect diffraction data sets optimised for maximum resolution with minimised radiation damage. In less than two months of operation more than 2.3 million diffraction images have been collected from 1422 samples ranging from initial hits from crystallisation experiments to large-scale data set collection for drug discovery programmes.<\/p>\n\n\n\n<div class=\"wp-block-image wp-image-2826 size-full\"><figure class=\"vf-figure  | vf-figure--align vf-figure--align-centered \"><img loading=\"lazy\" decoding=\"async\" width=\"620\" height=\"380\" class=\"vf-figure__image\" src=\"http:\/\/news.embl.de\/wp-content\/uploads\/2014\/11\/1411_MASSIF-1_new.jpg\" alt=\"1411_MASSIF-1_new\" class=\"wp-image-2826\" srcset=\"https:\/\/www.embl.org\/news\/wp-content\/uploads\/2014\/11\/1411_MASSIF-1_new.jpg 620w, https:\/\/www.embl.org\/news\/wp-content\/uploads\/2014\/11\/1411_MASSIF-1_new-300x184.jpg 300w\" sizes=\"auto, (max-width: 620px) 100vw, 620px\" \/><figcaption class=\"vf-figure__caption\">Technical overview of MASSIF 1. IMAGE: Matthew Bowler<\/figcaption><\/figure><\/div>\n\n\n\n<p>As MASSIF1 is fully automatic, data are collected for the first time in a consistent manner and should allow the accumulation and comparison of a large amount of information that was previously unknown, including the exact dimensions of crystals and deeper information about their quality. Once the beamline has been running for an extended period, it will provide a treasure trove of additional information to feed back into crystallisation experiments and the software used to collect the data.<\/p>\n\n\n\n<blockquote class=\"vf-blockquote\"><p>It will provide a treasure trove of additional information to feed back into crystallisation experiments and the software used to collect the data.<\/p><\/blockquote>\n\n\n\n<p>The automatic routines developed are often able to locate crystals more effectively than the human eye and in many cases have obtained higher resolution data sets as all positions within a sample can be evaluated for diffraction quality.
