{"id":1344,"date":"2014-08-06T13:21:05","date_gmt":"2014-08-06T11:21:05","guid":{"rendered":"http:\/\/news.embl.de\/?p=1344"},"modified":"2024-11-14T16:32:21","modified_gmt":"2024-11-14T15:32:21","slug":"unpacking-iron-overload","status":"publish","type":"post","link":"https:\/\/www.embl.org\/news\/science\/unpacking-iron-overload\/","title":{"rendered":"Unpacking iron overload"},"content":{"rendered":"\n<h3 class=\"wp-block-heading\">By Lena Raditsch<\/h3>\n\n\n\n<p>In a study published today in <em>Cell Metabolism<\/em>, the joint <a title=\"Hentze Group\" href=\"http:\/\/www.embl.de\/research\/units\/directors_research\/hentze\/\" target=\"_blank\" rel=\"noopener noreferrer\">research team of Matthias Hentze<\/a>, Director of EMBL, and Martina Muckenthaler, professor at the University of Heidelberg, has demonstrated for the first time in living organisms that, in a rare form of haemochromatosis caused by a small mutation in the protein Ferroportin, which transports iron, the organ most affected by the disease\u2019s characteristic iron overload is the pancreas.<\/p>\n\n\n\n<p>Sandro Altamura, scientist at University of Heidelberg, and Bruno Galy, EMBL scientist genetically engineered Ferroportin in mice so that they could selectively disrupt its interaction with Hepcidin, a hormone that counteracts Ferroportin\u2019s activity to maintain the balance of iron in the body. \u201cOur research now reveals that this rare form of haemochromatosis is caused by an erroneous signal cascade: Hepcidin is unable to switch Ferroportin off and so too much iron is taken up by the body and many of its organs,\u201d says Martina Muckenthaler.<\/p>\n\n\n\n<blockquote class=\"vf-blockquote\"><p>Our research now reveals that this rare form of haemochromatosis is caused by an erroneous signal cascade<\/p><\/blockquote>\n\n\n\n<p>\u201cThis animal model helps us to further understand the detrimental effects the excess of iron has, especially on the pancreas, which was unexpected. Ultimately, the mouse model can help to develop ideas for the treatment of iron overload diseases,\u201c says Matthias Hentze.<\/p>\n\n\n\n<p>The research was carried out within the joint <a title=\"Molecular Medicine Partnership Unit\" href=\"http:\/\/www.embl.de\/mmpu\/mmpu\/\" target=\"_blank\" rel=\"noopener noreferrer\">Molecular Medicine Partnership Unit,<\/a> a collaboration between EMBL and the University of Heidelberg.
