Our current research activities are focused on live cell studies of protein kinetics and (the dynamics of) protein-protein interactions in these processes. We use GFP-tagging of DNA-repair factors (e.g. ERCC1, XPA) and both general and hormone inducible transcription factors (eg. TFIIH, AR and coregulators), and study their functioning in living mammalian cells with confocal time-lapse imaging and quantitative fluorescence assays such as fluorescence recovery after photobleaching (FRAP), fluorescence resonance energy transfer (FRET) and fluorescence correlation spectroscopy (FCS). In addition, for optimal interpretation of the complex and massive amount of experimental results we develop and apply computer modeling methodology to simulate both FRAP and FRET assays as well as the behavior of proteins in repair and transcription.
