Hentze and Andreas E. Kulozik

Our research interest

Our team focuses on basic and translational aspects of RNA biology in cancer, with a particular focus on childhood cancers. Specific areas of interest include cell stress-induced changes in posttranscriptional gene regulation, altered behaviour of RNA-binding proteins (RBPs) in cancer, synthetic lethality relationships in cancer, the roles of RBPs in modulating the efficacy of existing chemotherapeutics, and drug repurposing efforts to bring our discoveries towards translational applications.
Our team using a diverse array of approaches, from classical molecular and cellular biochemistry techniques to state-of-the-art proteomic screening methods. Through further collaborations with the groups of Dr. Chris Carnie and Prof. Thomas Grünewald, we supplement our expertise with complementary genetic screening approaches and in vivo models of pediatric sarcomas.

Background

Initially, our group’s interest in RNA biology arose from detailed clinical analyses of two common hematological disorders, ß-thalassemia and thrombophilia, respectively. Interestingly, it is now becoming apparent that the principles that have been uncovered by the analysis of these two model conditions are applicable more generally.

Furthermore, the results of these projects have illustrated that the detailed analysis of unexplained medical phenomena can result in the identification and characterization of previously unrecognized basic biological mechanisms with broad implications.

Indeed, our recent work has highlighted a wealth of RBPs that exhibit differential RNA-binding in osteosarcoma, a childhood bone cancer with an urgent need for better treatment options.

Goals

Our driving aim is to build upon mechanistic discoveries that have the potential to both transform our understanding of fundamental biology and highlight promising new therapeutic strategies for childhood cancers including but not limited to osteosarcoma. Our unique partnership enables deep mechanistic characterisation of ‘disease-relevant’ RNA biology as well as a clear translational line-of-site towards identifying the next generation of therapeutic interventions.

Join us!

We are always keen to hear from others who share our interest in these fundamental areas of biology, and welcome speculative applications from prospective research assistants, master/doctoral students and postdocs.

<details class="vf-details" id="" >

Selected publications

Systematic analysis of RNA-binding proteins identifies targetable therapeutic vulnerabilities in osteosarcoma.
Zhou Y, Ray PS, Zhu J, Stein F, Rettel M, Sekaran T, Sahadevan S, Perez-Perri JI, Roth EK, Myklebost O, Meza-Zepeda LA, von Deimling A, Fu C, Brosig AN, Boye K, Nathrath M, Blattmann C, Lehner B, Hentze MW, Kulozik AE.
Nat Commun. 2024 Apr 1;15(1):2810. doi: 10.1038/s41467-024-47031-y.PMID: 38561347

Blasticidin S inhibits mammalian translation and enhances production of protein encoded by nonsense mRNA.
Powers KT, Stevenson-Jones F, Yadav SKN, Amthor B, Bufton JC, Borucu U, Shen D, Becker JP, Lavysh D, Hentze MW, Kulozik AE, Neu-Yilik G, Schaffitzel C.
Nucleic Acids Res. 2021 Jul 21;49(13):7665-7679. doi: 10.1093/nar/gkab532.
