For example, in structural biology insights into the functional mechanisms can be obtained by elucidating the 3D molecular structure of proteins and protein complexes using technologies such as X-ray crystallography, cryo-EM and NMR. In biochemistry and biophysics, interactions with other proteins, nucleic acids and small molecules can be studied by determining affinities and specificities. Proteins can function as antigens to generate specific antibodies or as reagents in cell biology experiments. Furthermore, recombinant proteins can be used as tool molecules in genomics, chemical biology and microscopy assays. In order for these experiments to produce reliable and biologically relevant results, they must be performed using high-quality proteins that are active, properly folded, in the right oligomeric state and contain correctly inserted co-factors. Ensuring that these parameters are fulfilled requires quality control. <\/p>\n\n\n\n<p>In this course, we want to help the participants to become acquainted with the concept of protein quality control and provide them with an overview of the specific protein requirements for various downstream applications. Different techniques that can be applied for this purpose will be introduced and participants will have the opportunity to get hands-on experience with these technologies.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Audience<\/h2>\n\n\n\n<p>This course is aimed at life-science researchers of all levels, who are using or plan to use purified proteins in their wet-lab experiments, but who do not necessarily have a strong background in protein production and characterisation methods. We will focus on introducing protein quality control methodologies in order to improve the reliability and reproducibility of data obtained from experiments that make use of purified proteins. After this course, participants should be familiar with the biochemical and biophysical techniques that can be applied to ensure that they are working with high-quality protein samples suitable for the planned downstream experiments.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Modules\/resources<\/h2>\n\n\n\n<ul class=\"wp-block-list\"><li>Isothermal Titration Calorimetry (ITC) and Dynamic Light Scattering (DLS)<\/li><li>Nano-Differential Scanning Fluorimetry (nano-DSF) and MicroScale Thermophoresis (MST)<\/li><li>Size Exclusion Chromatography coupled to Multi-Angle Light Scattering and Dynamic Light Scattering (SEC-MALS-DLS) and Mass Photometry<\/li><li>Cryo-Electron Microscopy (cryo-EM)<\/li><\/ul>\n\n\n\n<h2 class=\"wp-block-heading\">Learning outcomes<\/h2>\n\n\n\n<p>During the course, specialists from both academia and industry will work closely together to introduce technologies frequently used in protein quality control. Some of these methodologies can be utilised both to assess protein quality and as stand-alone biophysical characterisation techniques. <\/p>\n\n\n\n<p>One of the highlights of the course that we are very excited about is the cryo-electron microscopy (cryo-EM) practical. Cryo-EM has undergone a revolution in recent years and is becoming increasingly popular in many interdisciplinary projects. However, in order to attain high-resolution data, the protein samples need to fulfil quite stringent conditions.<\/p>\n\n\n\n<p>Our main goals are:<\/p>\n\n\n\n<ul class=\"wp-block-list\"><li>Teach the participants how to perform proper protein quality control checks<\/li><li>Introduce the participants to various biophysical techniques used for protein characterisation, such as SEC-MALS, DLS, mass photometry, nano-DSF, spectroscopy and mass spectrometry<\/li><li>Show participants how to characterise protein interactions using ITC and MST<\/li><li>Provide participants with a basic background on cryo-EM and show the effects sample quality has on the acquired data<\/li><li>Where possible, allow participants to apply these technologies onto their own protein samples in the practicals<\/li><li>Overall, increase the success rate, reliability and reproducibility of downstream experiments in which purified proteins are used<\/li><\/ul>\n<\/section>\n\n\n\n<section The grant is restricted to PhD students and postdocs who conduct basic biomedical research.<br>Whether you are eligible to apply for a travel grant,&nbsp;depends on when you received your university entrance qualification (e.g. Abitur, A-Levels, High School Diploma, Final State Examination):<\/p>\n\n\n\n<p>&#8211; for PhD and MD students, as well as graduates, the university entrance qualification must not have been obtained more than 11 years ago at the time of the envisaged course<br>&#8211; for postdocs, the university entrance qualification must not have been obtained more than 13 years ago at the time of the envisaged course<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Application<\/strong><\/h2>\n\n\n\n<p>You may apply for financial assistance when submitting your motivation letter for courses, and abstract for conferences. In your application, you will be asked to answer questions regarding why your lab cannot fund your attendance and how your attendance will make a difference to your career. Application for financial support will not affect the outcome of your registration application.<\/p>\n\n\n\n<p>For the Boehringer Ingelheim Fonds Travel Grant, there is a pre-application question during the motivation letter submission process, and if selected you will be requested to complete a standard form and documentation consisting of your travel expense estimation.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\"><strong>Selection<\/strong><\/h2>\n\n\n\n<p>The scientific organisers will select the recipients of all financial assistance during the motivation letter or abstract selection process.
