Please reach out to <a>events@embl.org<\/a> to request it.<\/p>\n\n\n\n<p>This conference will take place at EMBL Heidelberg, with the option to attend virtually.<\/p>\n\n\n\n<p><\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Symposium overview<\/h2>\n\n\n\n<p>In its second edition, this symposium will cover emerging key concepts of latent metastasis and dissemination of tumour cells including evolution and genetic diversity of metastasis, circulating tumour cells, stemness, dormancy and stromal reprogramming, transcriptional and epigenetic control, motility and invasive signalling, metastatic heterogeneity, EMT and plasticity, immune evasion, and metabolic adaptations. The meeting will provide a unique interdisciplinary exchange on current approaches and future collaborations on metastasis and its therapeutic challenges.<\/p>\n\n\n\n<p>Metastasis research is a nexus for several of the most exciting areas and technical developments of current cancer biology and translational molecular medicine. High-resolution intravital imaging, single-cell sequencing and integrated genomics approaches, innovative tissue modelling, and co-culture systems for disease screening and patient stratification all have enormous implications for our basic understanding of the mechanisms of this deadly disease and hold great promises for progress in its successful treatment. Engaging speakers, new science, and plenty of opportunity to network and socialise &#8211; all a stone&#8217;s throw from the centre of beautiful Heidelberg. With the rapid advancement of CRISPR\/Cas-based genome modification techniques in mice, we launched the Knock-Out All Project (KOAP) in 2018 to induce mutations in all protein coding genes within the mouse genome, using both conventional and conditional (floxed) methods. After five years of effort, we have achieved approximately 50% completion of the entire project, resulting in the establishment of the largest collection of genetically modified mouse strains. As of June 2024, we have generated and prepared for distribution a total of 10,881 knockout (KO) strains and 9,874 conditional knockout flox (cKO) strains.<\/p>\n\n\n\n<p>In order to facilitate engraftment of human cells, tissues, and organs without rejection by the functional immune system present in mice, various immune-compromised strains have been developed. To this end, we directly disrupted Prdkc and Il2rg genes in NOD mice to create an enhanced engraftment model known as NCG with improved acceptance of human immune cells and tissues. However, the NCG mouse models have limitations when it comes to supporting differentiation of immune cell populations other than T cells, such as myeloid cells and NK cells. To address this limitation, we are developing the next generation of NCG models with specific enhancements such as expression of human cytokines and HLA molecules along with increased engraftment capacity for human cells.<\/p>\n\n\n\n<p>These new models will be more suitable for studying differentiation of immune cell types and will be made available to academic researchers. The enhanced engraftment capability for human immune cells has been validated in our laboratory, and relevant data can be downloaded from our website when searching for specific
